dc.description.abstractอะแคนทามีบา (Acanthamoeba) เป็นโปรโตซัวชนิดเซลล์เดียวที่เคลื่อนที่ด้วยขาเทียม (pseudopodia) พบได้ทั่วไปในธรรมชาติ เช่น แหล่งน้ำ ดิน และฝุ่น เชื้อนี้สามารถก่อโรคในคนแบบติดเชื้อโดยบังเอิญ และเป็นสาเหตุสำคัญของ โรคกระจกตาอักเสบ (Acanthamoeba keratitis) และโรคสมองอักเสบแบบแกรนูโลมา (GAE) การศึกษานี้ดำเนินในพื้นที่ บึงโง้ง อำเภอเสาไห้ จังหวัดสระบุรี โดยตรวจหาเชื้อ Acanthamoeba spp. จากน้า จำนวน 30 ตัวอย่าง ด้วย 3 วิธี ได้แก่ การตรวจด้วยกล้องจุลทรรศน์ การเพาะเลี้ยง และการตรวจแบบ Conventional PCR พบว่า น้า มีอุณหภูมิเฉลี่ย 25.8°C และ pH 8.1 ผลตรวจด้วยกล้องจุลทรรศน์ไม่พบเชื้อ แต่ การเพาะเลี้ยงพบเชื้อร้อยละ 30 โดยมีลักษณะคล้ายกลุ่ม Astronyxid และ Polyphagid ซึ่งพบในสิ่งแวดล้อมและอาจก่อโรคในมนุษย์ ส่วนการตรวจด้วย PCR ไม่พบชิ้นส่วนดีเอ็นเอของเชื้อ อาจเกิดจากปริมาณดีเอ็นเอต่ำ หรือมีสารรบกวนสรุปได้ว่าการตรวจหาเชื้อ Acanthamoeba spp. ในแหล่งน้ำ ควรใช้ทั้งวิธีเพาะเลี้ยงและเทคนิคอณูชีววิทยาร่วมกัน เพื่อเพิ่มความแม่นยำ และความไวในการตรวจหาเชื้อจากสิ่งแวดล้อม  en_US

dc.description.other-abstractAcanthamoeba is a free-living unicellular protozoan that moves using pseudopodia. It is commonly found in natural environments such as water, soil, and dust. Human infection occurs accidentally and may lead to Acanthamoeba keratitis or granulomatous amoebic encephalitis (GAE). This study investigated water sources at Bueng Ngong, Sao Hai District, Saraburi Province, Thailand, using three detection methods for Acanthamoeba spp.: microscopic examination, culture, and conventional PCR. Thirty water samples were collected, with an average temperature of 25.8°C and pH 8.1 Microscopic examination did not detect the organism. However, 30% of samples were positive in culture, with isolates showing characteristics similar to the Astronyxid and Polyphagid groups, which are environmentally common and potentially pathogenic. Conventional PCR did not detect Acanthamoeba DNA, likely due to low DNA concentration or PCR inhibitors. In conclusion, detection of Acanthamoeba spp. in environmental water should combineculture and molecular methods to improve accuracy and sensitivity.en_US
