dc.description.abstractรายงานการวิจัย เรื่อง การพัฒนาตำรับอนุภาคนาโนสำหรับรักษาบาดแผลที่ผิวหนังจากสารสกัดเอทานอลของข้าวหักไรซ์เบอร์รี่ เป็นการวิจัยทางห้องปฏิบัติการ เพื่อพัฒนาทรานสเฟอร์โซมที่บรรจุสารสกัดจากข้าวหักไรซ์เบอร์รี่ เพื่อเพิ่มความคงตัวให้กับสารสกัด และเพิ่มฤทธิ์ในการสมานบาดแผลของสารสกัด ข้าวหักไรซ์เบอร์รี่เกิดขึ้นอย่างหลีกเลี่ยงไม่ได้จากกระบวนการสีข้าวอินทรีย์ในประเทศไทย ที่ผลิตได้หลายตันต่อปีและมีมูลค่าต่ำ มีงานวิจัยที่ตีพิมพ์ถึงสารอาหารอันทรงคุณค่าในสารสกัดข้าวหักไรซ์เบอร์รี่ (RBE) เช่น แกมมาโอไรซานอล ปริมาณกรดไขมัน สารฟีนอลิก และสารฟลาโวนอยด์ ที่มีฤทธิ์ต้านอนุมูลอิสระ สารอาหารเหล่านี้สามารถนำไปใช้เป็นส่วนประกอบสำคัญในผลิตภัณฑ์ยาได้ แต่ยังไม่มีการใช้ประโยชน์จากกลไกนี้ในการสมานบาดแผล ในงานวิจัยนี้ ได้มีการพยายามพัฒนาทรานสเฟอร์โซมที่บรรจุสารสกัดจากข้าวหักไรซ์เบอร์รี่ (T-RBE) เพื่อเพิ่มความคงตัวให้กับสารสกัด และเพิ่มฤทธิ์ทางชีวภาพของสารสกัด เทคโนโลยีสีเขียวถูกนำมาใช้ในการเตรียม T-RBE โดยเป็นรายแรกที่ใช้ลาโนลิน PEG-75 (สารลดแรงตึงผิวที่ไม่มีประจุ) ทำหน้าที่เป็นเอจแอคติเวเตอร์ ผ่านกระบวนการโฮโมจิไนเซชันด้วยความเร็วสูง โดยไม่ต้องใช้ตัวทำละลายอินทรีย์ ส่งผลให้ได้อนุภาคทรงกลมขนาด 256.60 ± 1.59 นาโนเมตร ศักย์ซีตาเชิงลบที่ 35.30 ± 1.26 ค่าดัชนีการกระจายตัวของอนุภาคต่ำกว่าหนึ่ง (0.25 ± 0.01 mV) และประสิทธิภาพการกักเก็บสารสำคัญคือ 65.30 ± 2.98% โดย T-RBE มีความคงตัวสูงในช่วงเวลา 3 เดือน ไม่เป็นพิษต่อเซลล์ไฟโบรบลาสต์และเซลล์ HaCaT ของมนุษย์ และสามารถเพิ่มการสมานบาดแผล โดยผ่านเส้นทางการส่งสัญญาณ FAK/AKT ที่เกี่ยวกับการเคลื่อนที่ของเซลล์ ลดระดับการผลิตไนไตรต์ และยับยั้งการเติบโตของเชื้อ S. aureus โดยเทียบกับสารสกัด สูตรตำรับ T-RBE ช่วยให้การปลดปล่อยสารสำคัญเป็นแบบต่อเนื่อง และเพิ่มการซึมผ่านสารสำคัญผ่านผิวหนังได้อย่างมีประสิทธิภาพ เหนือกว่าสารสกัด ซึ่งชี้ให้เห็นถึงศักยภาพในการใช้ T-RBE ในระบบผิวหนังสำหรับการรักษาบาดแผลที่บกพร่องและหายช้าen_US

dc.description.other-abstractThe research entitles “Development of nanoparticle formulation for skin wound healing from ethanolic riceberry broken rice extract” is the in-vitro study to develop riceberry broken rice extract-loaded transfersomes for improving the extract stability, and subsequently enhanced its wound healing property. Riceberry broken rice is inevitably generated from milling organic rice process in Thailand for several tons per year with low value. The valuable nutrients of Riceberry broken rice extract (RBE) has been published such as gamma-oryzanol, fatty acid content, phenolics and flavonoids with antioxidant activities. These can be applied as potentially active ingredients in pharmaceutical products, but its mechanisms for wound healing have not been exploited. In this research, an attempt has been made to develop RBE-loaded transfersomes (T-RBE) for improving the extract stability, and subsequently enhanced its bioactivities. The green technology was used to prepare T-RBE with the first to use PEG-75 lanolin (nonionic surfactant) as edge activator via a high speed homogenization process without the use of organic solvents, resulting in homogeneous spherical nanoparticles with sizes of 256.60 ± 1.59 nm, negative zeta potential in -35.30 ± 1.26, polydispersity index value less than one (0.25 ± 0.01 mV), and entrapment efficiency of 65.30 ± 2.98 %. The TRBE was highly stable over a period of 3 months, not toxic to human fibroblast and HaCaT cells, and was able to enhance wound healing via the FAK/AKT signaling pathway, decrease nitrite production levels and inhibit the growth of S. aureus compared to the extract. TRBE formulation effectively sustained release of biological compounds and efficiently enhanced skin penetration over the extract, suggesting a potential use of T-RBE as dermal systems for impaired and delayed wound treatmenten_US
